Gene Expression

cDNA library

  • Construction and validation of 2 cDNA libraries from M. acuminata 'Calcutta 4' and 'Cavendish' constituted from leaf fragments infected in vitro by M. fijiensis 27648 clones (CIRAD)
  • Development and characterization of cDNA libraries from M. acuminata 'Calcutta 4' (leaves from plants submitted to stresses of temperature, in vitro leaves infected with M. fijiensis , male flower, roots from in vitro plants, and peel from green fruits) (EMBRAPA)
  • Construction of substraction libraries of two types: One made from ripe fruit (stages 5 and 7) and the other from banana tissue infected with M. fijiensis at different times (CINVESTAV)
  • Construction of SSH cDNA libraries to study BSV-Ol activation through in-vitro culture of plantain landrace 'Penkelon noir' (CIRAD)
  • Construction of a suppression subtractive hybridization (SSH)-cDNA library for Cavendish bananas (FABI)
  • cDNA library construction from M. balbisiana (B genome), and development of EST sequence data (IITA)
  • Representation Difference Analysis (RDA) to isolate markers and or genes related to wounding and/or banana weevil infestation. From this activity a cDNA library has been generated (NARO, FABI)

EST clustering and annotation

  • Identification and characterisation of genes involved in fungal resistance (UOM, EMBRAPA)
  • Molecular and phylogenetic analysis of the ribulose 1,5 bisphosphate carboxylase gene family (UQ)
  • Isolation and identification of ripening-related genes involved in sugar metabolism, and initiation and regulation of ripening banana process throughout SSH cDNA libraries and RT-PCR approaches (CIRAD)
  • Characterization of Resistance Genes Analogs (RGAs) from M. balbisiana 'Calcutta 4' (EMBRAPA)
  • Sequence analysis of EST clones from 'Pisang Rastali' and variant (UM)

Microarrays and gene expression profiling

  • Spotting of the cDNA library on micro-array slides (FABI)
  • Microarray experiments (UM)
  • Comparative analysis of the transcriptome during interactions of plants with pathogenic, non-pathogenic, beneficial and mycorrhizal fungi (UQ)

SAGE

  • Adaptation of SuperSAGE and a global analysis of the banana transcriptome (KUL)
